Biochemical characteristics of sperm from Siberian sturgeon (Acipenser baerii) farmed in Vietnam

Abstract: This study aimed to evaluate the sperm quality parameters of Siberian sturgeon (Acipenser baerii) reared in highland conditions in Lam Dong Province, Vietnam-a region with a distinct temperate climate that is highly suitable for the cultivation of this cold-water species. Semen was collected from four mature male individuals and analyzed for key physiological and biochemical parameters, including semen volume, sperm density, spermatocrit rate, sperm motility percentage and duration, as well as the ionic and protein composition of the seminal plasma. The results showed that the average semen volume reached 51.30 mL per individual, with a sperm density of 1.78 × 10⁹ cells/mL and a spermatocrit value of 53.80%, indicating a high cellular concentration. The average sperm motility rate was 67.5%, with a motility duration of approximately 152.3 seconds. Seminal plasma analysis revealed the presence of essential ions such as sodium (32.96 mM), potassium (3.11 mM), calcium (0.27 mM), magnesium (0.36 mM), and chloride (10.88 mM), along with a total protein content of 0.56 mg/mL and an osmolality of 76.10 mOsm/kg. These parameters indicate that the sperm of Siberian sturgeon (A. baerii) cultured in tropical highland conditions retains good reproductive quality and fully meets the requirements for artificial insemination. This is the first research in Vietnam to provide a comprehensive data on the sperm quality of A. baerii under highland aquaculture conditions, contributing scientific foundations for the optimization of artificial propagation techniques and sperm cryopreservation conditions adapted to local environments.
